Just through playing SSE normally... you should unlock the following.
In order of unlocking:
Marth -> Falco -> Captain Falcon -> Lucario -> Snake -> Mr. Game and Watch -> R.O.B -> Luigi -> Ness -> Ganondorf (requires recollection of Ling and Zelda in SSE) -> Sonic
After clearing the SSE, if you revisit the ruins, you can find Wolf, visiting the forest will let you find toon link and the swamp-ish area will let you find jigglypuff.
Non-SSE Method
------------------------------------
Toon Link - Clear classic mode (after beating SSE)
Luigi - Clear classic mode with easy difficulty
Captain Falcon - Clear classic mode with normal difficulty
Marth - Clear classic mode with 3 people
Ganondorf - Clear classic mode on hard difficulty with Link and Zelda/Sheik. (Continues allowed)
Falco - Multiman Melee (100 man melee)
Lucario - Beat Break the targets level 1 to 5
Ness - Reflect 10 shots
Jigglypuff - After clearing SSE, beat event number 20
Sonic - Beat classic mode with 10 characters
Snake - Play on shadow moses island for 15 times
Mr Game and Watch - Beat break the target level 1 with over 30 characters
Wolf - After beating SSE, use fox and beat 'Boss Battle' mode
Through Brawling (VS mode)

Lv2 Beat classic mode with easy TWICE, or beat it with normal ONCE.
Lv3 Beat classic mode with normal TWICE, or beat it with hard ONCE.
Lv4 Beat classic mode with hard TWICE, or beat it with very hard ONCE
Lv5 Beat classic mode with very hard TWICE.
Events
Missions 21 - (28 to 30) Beat events 1 - 20 and have R.O.B unlocked
Missions (29 to 31) - 40 Beat all the remaining events and have everyone unlocked
Mission 41 - Beat mission 40

So we have to beat ALL the characters break the targets lvls 1-5 to unlock him?
*crag* that I'm going through SSE unless it's just 1 entire break the target thing where you can pick any character to do it. I change my mind, going through Classic about 4 times will probably be longer than going through SSE enough to Lucario |

Sonic sounds too easy to unlock.
You can just beat Classic Mode with 10 characters on Easy, Normal, And Hard, beat the crap out of everybody, and you unlock Luigi, Falcon, Ganondorf, Marth and Sonic in the process. |

Maybe they switched the requirements of the two because Luigi was getting an even bigger fan base? |
