From what I've seen in screenshots, it seems to be about an occurence causing toads to become giant spherical fatties,
Presumably, Fawful is the main antagonist this time. Not just because of the part with bowser, but there's also this screenshot.
And to top it all off, you go inside bowser's stomache. It looks like you control Bowser to affect things in his stomache so the Marios can go through. Why did he eat them in the first place, anyways?
I hate to say it, but Commander shroob is right. Those saucers are green, and therefore probably belong to fawful.
