It was confirmed on October 2nd at Nintendo's 2008 Fall Conference and is slated for a 2009 release in Japan.
Apparently, it features Bros. Items again, as well as the ability to play as Bowser in some parts, while Mario and Luigi try to do something about...er, Toads becoming giant? o.O But what REALLY has me excited is the apparent role as Fawful as either the main or at least a minor villain.
In this screen, Bowser chases a flying red and green figure, thought to be either Fawful or Prince Peasely.
And, well...Look at those monsters! They're wearing Fawful masks! O:
If I had to make a rough theory about the storyline, I'd wager that Fawful tries to take over the Mushroom Kingdom by spreading some kind of enlarging disease through the water from his hideout in the sewers...But because so little has been released, no one can be sure.
Sorry, but Fawful just isn't "villain material". He has always been "hilarious henchman material" and if Nintendo changes that then M'aiq will lose even more respect for them. As awesome as Fawful is, what the hell would he do in the major villain limelight?

Just because he speaks with fury while talking about sandwich related items mean he's not suitable. His Mustard of Doom is the first step in the Sandwich of world domination. I think he would be a good villain for the Fink-Rats to fight.
This looks great. It's great to see Mario and Luigi's next RPG, and it will certainly be interesting to see Fawful as the main villain. I think it will suit the game fairly well. I can't wait to see more information about it. There are just 3 things I noticed about it, though...
